研究の概要

 感染症学研究室の理念は、science for society、即ち、社会(一般市民あるいは感染症患者)のための科学を実践することである。その実現のために、研究室と社会あるいは医療施設(大学病院?基幹病院?クリニックなど)との連携を大切にしている。現場から頂いた感染症に関する課題?問題点への対処や解決に結び付くようなヒントを連携者へ提示するよう心掛けている。具体的には、呼吸器感染症?皮膚軟部組織感染症?人獣共通感染症?セプシスといった様々領域の疾患や医療施設内での感染管理に関する研究を行っている。以下に挙げる6項目が主たる研究テーマである。くわえて、国際学生交流(大韓民国?慶尚国立大学医学部と北里生命科学研究所)も実践している。

研究の概要

  1. ヘリコバクター?ハイルマニイ感染症に関する迅速診断?除菌、感染予防技術の開発
    ハイルマニイの感染診断には、胃生検から調製したDNAに含まれるハイルマニイの16S rRNA遺伝子をPCRで検出する方法が用いられている。しかし、内視鏡による生検は大変である上に、菌が寄生する場所を採取しなければならないという問題がある。そこで、ピロリと同時の感染診断を可能とする、血液?尿(ハイルマニイ抗体の検出)、便(組織化学にも使用可能なハイルマニイVHH抗体の利用)を検体とした迅速検査法を開発している。除菌および感染予防においては、ハイルマニイの薬剤耐性に関わる遺伝子解析、狭域スペクトルの新規抗ヘリコバクター剤の開発、乳酸菌による新たな感染防御システムの開発を行っている。
  2. A群連鎖球菌(group A streptococci, GAS)のM蛋白質の受容体であるヒトCD46を発現するCD46トランスジェニックマウスを用いた、急激な骨破壊機構の解析
    皮膚および軟部組織から感染したGASは、血液に侵入後に骨髄内の血管を破壊し滲出する。線維芽細胞を含む骨髄内の細胞表層のCD46にGASが付着し、その刺激により大量のRANKLが発現する。骨髄中の破骨前駆細胞はRANKLにより活性化破骨細胞に分化し、海綿骨から皮質骨に向かって急激な骨吸収を生じる。従ってRANKL抗体の投与により、誘導される骨吸収は抑制できる。
  3. Salmonella enterica serovar Typhimuriumの感染により誘導される免疫応答の解析
    マウスを用いた感染実験などに最適のサルモネラの野生株や各種遺伝子変異株を提供している。また、遺伝子変異や感染実験の助言も行っている。
